SPPS stands for Solid-Phase Peptide Synthesis, which is a technique used in organic chemistry and biochemistry to synthesize peptides, which are short chains of amino acids. It is a widely adopted method for synthesizing peptides due to its efficiency and precision.
In SPPS, the process begins by anchoring the first amino acid onto a solid-phase support such as polystyrene resin. This starting amino acid is typically protected to prevent undesired reactions. Subsequently, each amino acid is sequentially added onto the growing peptide chain using a coupling reagent. This step involves removing the protective group on the newly added amino acid and chemically bonding it to the growing chain. The process is repeated until the desired peptide sequence is achieved.
The solid-phase support allows for easy purification and isolation of the desired peptide product. Unbound reagents and impurities can be efficiently washed away while the anchored peptide remains attached to the resin. After synthesis, the peptide is cleaved from the solid support, resulting in a crude peptide mixture that may undergo further purification steps if necessary.
